Research-grade semaglutide is supplied as a lyophilized (freeze-dried) powder. Before it can be used in experimental protocols, it must be reconstituted by dissolving it in an appropriate solvent. The reconstitution process requires sterile technique and accurate measurements to ensure the resulting solution has the intended concentration.
This guide is written for researchers working with semaglutide as a research compound in a laboratory setting. It is not a guide to pharmaceutical semaglutide preparation or self-injection.
What You Need
| Parameter | Detail |
| Lyophilized semaglutide vial | Research-grade, verified by HPLC and mass spectrometry |
| Bacteriostatic water (BAC water) | 0.9% benzyl alcohol in sterile water; preferred for extended storage stability |
| Sterile insulin syringe (1 mL) | For precise measurement of small volumes |
| Alcohol swabs (70% isopropanol) | For sterilising vial septums before puncture |
| Clean, flat workspace | Ideally within a laminar flow hood for maximum sterility |
Calculating the Desired Concentration
Before reconstitution, decide on the target concentration of the resulting solution. Common research concentrations for semaglutide range from 0.5 mg/mL to 2 mg/mL, depending on the intended dose volumes.
The calculation is straightforward: divide the total peptide mass (in mg) by the target concentration (in mg/mL) to determine the volume of solvent to add.
Example: A 5 mg vial of semaglutide reconstituted with 2.5 mL of BAC water gives a concentration of 2 mg/mL. Each 0.1 mL drawn from this solution contains 0.2 mg of semaglutide. A 5 mg vial reconstituted with 5 mL gives 1 mg/mL, with each 0.1 mL containing 0.1 mg.
Step-by-Step Reconstitution Process
Step 1: Allow the semaglutide vial to reach room temperature before opening. Cold peptide powder can behave differently during dissolution.
Step 2: Wipe the rubber septum of the semaglutide vial and the BAC water vial with an alcohol swab. Allow to air dry for 30 seconds.
Step 3: Draw the calculated volume of BAC water into a sterile syringe. Introduce the needle slowly through the centre of the semaglutide vial septum.
Step 4: Direct the stream of BAC water slowly down the inside wall of the vial — not directly onto the powder. This minimises agitation and reduces the risk of denaturation.
Step 5: Once all the solvent has been added, gently swirl the vial in a circular motion. Do not shake. Semaglutide contains a fatty acid chain that can aggregate if vigorously agitated.
Step 6: If the powder does not dissolve immediately, allow the vial to sit at room temperature for 5–10 minutes, then gently swirl again. A clear, colourless to slightly yellow solution indicates successful dissolution.
Step 7: Inspect the solution. It should be clear without visible particulates. Cloudiness, visible flakes, or unusual colour indicate a problem — discard and start again.
Storage After Reconstitution
| Parameter | Detail |
| Storage location | Refrigerator (+4°C), upright, in the original vial or amber storage vial |
| Maximum storage duration | 28 days with bacteriostatic water; 7 days with plain sterile water |
| Light exposure | Minimise — store in amber vial or in the dark within the refrigerator |
| Freezing reconstituted solution | Not recommended — fatty acid chain may aggregate on freeze-thaw |
| Before each use | Visually inspect for clarity and particulates; swirl gently if any settling observed |
Common Reconstitution Errors
Adding solvent too quickly: Introducing a large volume of solvent directly onto the powder can cause physical disruption of the lyophilized cake. Slow, wall-directed addition produces a more consistent result.
Using the wrong solvent: Plain sterile water can be used for immediate use, but bacteriostatic water is strongly preferred for any solution that will be stored. The benzyl alcohol preservative prevents bacterial contamination.
Shaking vigorously: The fatty acid chain on semaglutide can form aggregates if the solution is shaken. Always use gentle swirling.
Incorrect concentration calculation: Double-check the calculation before adding solvent. An error at this step propagates through all subsequent dose measurements.
